17. What Do I Look Like? The Wizard Of Oz? You Need A Brain? You Need A Heart? Go Ahead.Take Mine. Take Everything I Have. HEART? GO AHEAD. TAKE MINE. TAKE EVERYTHING I HAVE. I sort of had a plan as I ran to the Cullens' garage. The second part of it was totaling the bloodsucker's car on my way back. So I was at a loss when I mashed the button on the keyless remote, and it was not his Volvo that beeped and flashed its lights for me. It was another car – a standout even in the long line of vehicles that were mostly all drool-worthy in their own ways. Did he actually mean to give me the keys to an Aston Martin Vanquish, or was that an accident? I didn't pause to think about it, or if this would change that second part of my plan. I just threw myself into the silky leather seat and cranked the engine while my knees were still crunched up under the steering wheel. The sound of the motor's purr might have made me moan another day, but right now it was all I could do to concentrate enough to put it in drive. I found the seat release and shoved myself back as my foot rammed the pedal down. The car felt almost airborne as it leaped forward. It only took seconds to race through the tight, winding drive. The car responded to me like my thoughts were steering rather than my hands. As I blew out of the green tunnel and onto the highway, I caught a fleeting glimpse of Leah's gray face peering uneasily through the ferns. For half a second, I wondered what she'd think, and then I realized that I didn't care. I turned south, because I had no patience today for ferries or traffic or anything else that meant I might have to lift my foot off the pedal. In a sick way, it was my lucky day. If by lucky you meant taking a well-traveled highway at two hundred without so much as seeing one cop, even in the thirty-mile-an-hour speed-trap towns. What a letdown. A little chase action might have been nice, not to mention that the license plate info would bring the heat down on the leech. Sure, he'd buy his way out of it, but it might have been just a little inconvenient for him. The only sign of surveillance i came across was just a hint of dark brown fur flitting through the woods, running parallel to me for a few miles on the south side of Forks. Quil, it looked like. He must have seen me, too, because he disappeared after a minute without raising an alarm. Again, I almost wondered what his story would be before I remembered that I didn't care. I raced around the long U-shaped highway, heading for the biggest city I could find. That was the first part of my plan. It seemed to take forever, probably because I was still on the razor blades, but it actually didn't even take two hours before I was driving north into the undefined sprawl that was part Tacoma and part Seattle. I slowed down then, because I really wasn't trying to kill any innocent bystanders. This was a stupid plan. It wasn't going to work. But, as I'd searched my head for any way at all to get away from the pain, what Leah'd said today had popped in there. That would go away, you know, if you imprinted. You wouldn't have to hurt over her anymore. Seemed like maybe getting your choices taken away from you wasn't the very worst thing in the world. Maybe feeling like this was the very worst thing in the world. But I'd seen all the girls in La Push and up on the Makah rez and in Forks. I needed a wider hunting range. So how do you look for a random soul mate in a crowd? Well, first, I needed a crowd. So I tooled around, looking for a likely spot. I passed a couple of malls, which probably would've been pretty good places to find girls my age, but I couldn't make myself stop. Did I want to imprint on some girl who hung out in a mall all day? I kept going north, and it got more and more crowded. Eventually, I found a big park full of kids and families and skateboards and bikes and kites and picnics and the whole bit. I hadn't noticed till now – it was a nice day. Sun and all that. People were out celebrating the blue sky. I parked across two handicapped spots – just begging for a ticket – and joined the crowd. I walked around for what felt like hours. Long enough that the sun changed sides in the sky. I stared into the face of every girl who passed anywhere near me, making myself really look, noticing who was pretty and who had blue eyes and who looked good in braces and who had way too much makeup on. I tried to find something interesting about each face, so that I would know for sure that I'd really tried. Things like: This one had a really straight nose; that one should pull her hair out of her eyes; this one could do lipstick ads if the rest of her face was as perfect as her mouth___ Sometimes they stared back. Sometimes they looked scared – like they were thinking, Who is this big freak glaring at me? Sometimes I thought they looked kind of interested, but maybe that was just my ego running wild. Either way, nothing. Even when I met the eyes of the girl who was – no contest – the hottest girl in the park and probably in the city, and she stared right back with a speculation that looked like interest, I felt nothing. Just the same desperate drive to find a way out of the pain. As time went on, I started noticing all the wrong things. Bella things. This one's hair was the same color. That one's eyes were sort of shaped the same. This one's cheekbones cut across her face in just the same way. That one had the same little crease between her eyes – which made me wonder what she was worrying about___ That was when I gave up. Because it was beyond stupid to think that I had picked exactly the right place and time and I was going to simply walk into my soul mate just because I was so desperate to. It wouldn't make sense to find her here, anyway. If Sam was right, the best place to find my genetic match would be in La Push. And, clearly, no one there fit the bill. If Billy was right, then who knew? What made for a stronger wolf? I wandered back to the car and then slumped against the hood and played with the keys. Maybe I was what Leah thought she was. Some kind of dead end that shouldn't be passed on to another generation. Or maybe it was just that my life was a big, cruel joke, and there was no escape from the punch line. â€Å"Hey, you okay? Hello? You there, with the stolen car.† It took me a second to realize that the voice was talking to me, and then another second to decide to raise my head. A familiar-looking girl was staring at me, her expression kind of anxious. I knew why I recognized her face – I'd already catalogued this one. Light red-gold hair, fair skin, a few gold-colored freckles sprinkled across her cheeks and nose, and eyes the color of cinnamon. â€Å"If you're feeling that remorseful over boosting the car,† she said, smiling so that a dimple popped out in her chin, â€Å"you could always turn yourself in.† â€Å"It's borrowed, not stolen,† I snapped. My voice sounded horrible – like I'd been crying or something. Embarrassing. â€Å"Sure, thatW hold up in court.† I glowered. â€Å"You need something?† â€Å"Not really. I was kidding about the car, you know. It's just that†¦ you look really upset about something. Oh, hey, I'm Lizzie.† She held out her hand. I looked at it until she let it fall. â€Å"Anyway†¦,† she said awkwardly, â€Å"I was just wondering if I could help. Seemed like you were looking for someone before.† She gestured toward the park and shrugged. â€Å"Yeah.† She waited. I sighed. â€Å"I don't need any help. She's not here.† â€Å"Oh. Sorry.† â€Å"Me, too,† I muttered. I looked at the girl again. Lizzie. She was pretty. Nice enough to try to help a grouchy stranger who must seem nuts. Why couldn't she be the one? Why did everything have to be so freaking complicated? Nice girl, pretty, and sort of funny. Why not? â€Å"This is a beautiful car,† she said. â€Å"It's really a shame they're not making them anymore. I mean, the Vantage's body styling is gorgeous, too, but there's just something about the Vanquish___† Nice girl who knew cars. Wow. I stared at her face harder, wishing I knew how to make it work. C'mon, Jake – imprint already. â€Å"How's it drive?† she asked. â€Å"Like you wouldn't believe,† I told her. She grinned her one-dimple smile, clearly pleased to have dragged a halfway civil response out of me, and I gave her a reluctant smile back. But her smile did nothing about the sharp, cutting blades that raked up and down my body. No matter how much I wanted it to, my life was not going to come together like that. I wasn't in that healthier place where Leah was headed. I wasn't going to be able to fall in love like a normal person. Not when I was bleeding over someone else. Maybe – if it was ten years from now and Bella's heart was long dead and I'd hauled myself through the whole grieving process and come out in one piece again – maybe then I could offer Lizzie a ride in a fast car and talk makes and models and get to know something about her and see if I liked her as a person. But that wasn't going to happen now. Magic wasn't going to save me. I was just going to have to take the torture like a man. Suck it up. Lizzie waited, maybe hoping I was going to offer her that ride. Or maybe not. â€Å"I'd better get this car back to the guy I borrowed it from,† I muttered. She smiled again. â€Å"Glad to hear you're going straight.† â€Å"Yeah, you convinced me.† She watched me get in the car, still sort of concerned. I probably looked like someone who was about to drive off a cliff. Which maybe I would've, if that kind of move'd work for a werewolf. She waved once, her eyes trailing after the car. At first, I drove more sanely on the way back. I wasn't in a rush. I didn't want to go where I was going. Back to that house, back to that forest. Back to the pain I'd run from. Back to being absolutely alone with it. Okay, that was melodramatic. I wouldn't be all alone, but that was a bad thing. Leah and Seth would have to suffer with me. I was glad Seth wouldn't have to suffer long. Kid didn't deserve to have his peace of mind ruined. Leah didn't, either, but at least it was something she understood. Nothing new about pain for Leah. I sighed big as I thought about what Leah wanted from me, because I knew now that she was going to get it. I was still pissed at her, but I couldn't ignore the fact that i could make her life easier. And – now that I knew her better – I thought she would probably do this for me, if our positions were reversed. It would be interesting, at the very least, and strange, too, to have Leah as a companion – as a friend. We were going to get under each other's skin a lot, that was for sure. She wouldn't be one to let me wallow, but I thought that was a good thing. I'd probably need someone to kick my butt now and then. But when it came right down to it, she was really the only friend who had any chance of understanding what I was going through now. I thought of the hunt this morning, and how close our minds had been for that one moment in time. It hadn't been a bad thing. Different. A little scary, a little awkward. But also nice in a weird way. I didn't have to be all alone. And I knew Leah was strong enough to face with me the months that were coming. Months and years. It made me tired to think about it. I felt like I was staring out across an ocean that I was going to have to swim from shore to shore before I could rest again. So much time coming, and then so little time before it started. Before I was flung into that ocean. Three and a half more days, and here I was, wasting that little bit of time I had. I started driving too fast again. I saw Sam and Jared, one on either side of the road like sentinels, as I raced up the road toward Forks. They were well hidden in the thick branches, but I was expecting them, and I knew what to look for. I nodded as I blew past them, not bothering to wonder what they made of my day trip. I nodded to Leah and Seth, too, as I cruised up the Cullens' driveway. It was starting to get dark, and the clouds were thick on this side of the sound, but I saw their eyes glitter in the glow of the headlights. I would explain to them later. There'd be plenty of time for that. It was a surprise to find Edward waiting for me in the garage. I hadn't seen him away from Bella in days. I could tell from his face that nothing bad had happened to her. In fact, he looked more peaceful than before. My stomach tightened as I remembered where that peace came from. It was too bad that – with all my brooding – I'd forgotten to wreck the car. Oh well. I probably wouldn't have been able to stand hurting this car, anyway. Maybe he'd guessed as much, and that's why he'd lent it to me in the first place. â€Å"A few things, Jacob,† he said as soon as I cut the engine. I took a deep breath and held it for a minute. Then, slowly, I got out of the car and threw the keys to him. â€Å"Thanks for the loan,† I said sourly. Apparently, it would have to be repaid. â€Å"What do you want now?† â€Å"Firstly†¦ I know how averse you are to using your authority with your pack, but†¦Ã¢â‚¬  I blinked, astonished that he would even dream of starting in on this one. â€Å"What?† â€Å"If you can't or won't control Leah, then I – â€Å" â€Å"Leah?† I interrupted, speaking through my teeth. â€Å"What happened?† Edward's face was hard. â€Å"She came up to see why you'd left so abruptly. I tried to explain. I suppose it might not have come out right.† â€Å"What did she do?† â€Å"She phased to her human form and – â€Å" â€Å"Really?† I interrupted again, shocked this time. I couldn't process that. Leah letting her guard down right in the mouth of the enemy's lair? â€Å"She wanted to†¦ speak to Bella.† â€Å"To Bella?† Edward got all hissy then. â€Å"I won't let Bella be upset like that again. I don't care how justified Leah thinks she is! I didn't hurt her – of course I wouldn't – but I'll throw her out of the house if it happens again. I'll launch her right across the river – â€Å" â€Å"Hold on. What did she say?† None of this was making any sense. Edward took a deep breath, composing himself. â€Å"Leah was unnecessarily harsh. I'm not going to pretend that I understand why Bella is unable to let go of you, but I do know that she does not behave this way to hurt you. She suffers a great deal over the pain she's inflicting on you, and on me, by asking you to stay. What Leah said was uncalled for. Bella's been crying – â€Å" â€Å"Wait – Leah was yelling at Bella about me?† He nodded one sharp nod. â€Å"You were quite vehemently championed.† Whoa. â€Å"I didn't ask her to do that.† â€Å"I know.† I rolled my eyes. Of course he knew. He knew everything. But that was really something about Leah. Who would have believed it? Leah walking into the bloodsuckers' place human to complain about how /was being treated. â€Å"I can't promise to control Leah,† I told him. â€Å"I won't do that. But I'll talk to her, okay? And I don't think there'll be a repeat. Leah's not one to hold back, so she probably got it all off her chest today.† â€Å"I would say so.† â€Å"Anyway, I'll talk to Bella about it, too. She doesn't need to feel bad. This one's on me.† â€Å"I already told her that.† â€Å"Of course you did. Is she okay?† â€Å"She's sleeping now. Rose is with her.† So the psycho was â€Å"Rose† now. He'd completely crossed over to the dark side. He ignored that thought, continuing with a more complete answer to my question. â€Å"She's†¦ better in some ways. Aside from Leah's tirade and the resulting guilt.† Better. Because Edward was hearing the monster and everything was all lovey-dovey now. Fantastic. â€Å"It's a bit more than that,† he murmured. â€Å"Now that I can make out the child's thoughts, it's apparent that he or she has remarkably developed mental facilities. He can understand us, to an extent.† My mouth fell open. â€Å"Are you serious?† â€Å"Yes. He seems to have a vague sense of what hurts her now. He's trying to avoid that, as much as possible. He†¦ loves her. Already.† I stared at Edward, feeling sort of like my eyes might pop out of their sockets. Underneath that disbelief, I could see right away that this was the critical factor. This was what had changed Edward – that the monster had convinced him of this love. He couldn't hate what loved Bella. It was probably why he couldn't hate me, either. There was a big difference, though. I wasn't killing her. Edward went on, acting like he hadn't heard all that. â€Å"The progress, I believe, is more than we'd judged. When Carlisle returns – â€Å" â€Å"They're not back?† I cut in sharply. I thought of Sam and Jared, watching the road. Would they get curious as to what was going on? â€Å"Alice and Jasper are. Carlisle sent all the blood he was able to acquire, but it wasn't as much as he was hoping for – Bella will use up this supply in another day the way her appetite has grown. Carlisle stayed to try another source. I don't think that's necessary now, but he wants to be covered for any eventuality.† â€Å"Why isn't it necessary? If she needs more?† I could tell he was watching and listening to my reaction carefully as he explained. Tm trying to persuade Carlisle to deliver the baby as soon as he is back.† â€Å"What?† â€Å"The child seems to be attempting to avoid rough movements, but it's difficult. He's become too big. It's madness to wait, when he's clearly developed beyond what Carlisle had guessed. Bella's too fragile to delay.† I kept getting my legs knocked out from under me. First, counting on Edward's hatred of the thing so much. Now, I'd realized that I thought of those four days as a sure thing. I'd banked on them. The endless ocean of grief that waited stretched out before me. I tried to catch my breath. Edward waited. I stared at his face while I recovered, recognizing another change there. â€Å"You think she's going to make it,† I whispered. â€Å"Yes. That was the other thing I wanted to talk to you about.† I couldn't say anything. After a minute, he went on. â€Å"Yes,† he said again. â€Å"Waiting, as we have been, for the child to be ready, that was insanely dangerous. At any moment it could have been too late. But if we're proactive about this, if we act quickly, I see no reason why it should not go well. Knowing the child's mind is unbelievably helpful. Thankfully, Bella and Rose agree with me. Now that I've convinced them it's safe for the child if we proceed, there's nothing to keep this from working.† â€Å"When will Carlisle be back?† I asked, still whispering. I hadn't got my breath back yet. â€Å"By noon tomorrow.† My knees buckled. I had to grab the car to hold myself up. Edward reached out like he was offering support, but then he thought better of it and dropped his hands. â€Å"I'm sorry,† he whispered. â€Å"I am truly sorry for the pain this causes you, Jacob. Though you hate me, I must admit that I don't feel the same about you. I think of you as a†¦ a brother in many ways. A comrade in arms, at the very least. I regret your suffering more than you realize. But Bella is going to survive† – when he said that his voice was fierce, even violent – â€Å"and I know that's what really matters to you.† He was probably right. It was hard to tell. My head was spinning. â€Å"So I hate to do this now, while you're already dealing with too much, but, clearly, there is little time. I have to ask you for something – to beg, if I must.† â€Å"I don't have anything left,† I choked out. He lifted his hand again, as if to put it on my shoulder, but then let it drop like before and sighed. â€Å"I know how much you have given,† he said quietly. â€Å"But this is something you do have, and only you. I'm asking this of the true Alpha, Jacob. I'm asking this of Ephraim's heir.† I was way past being able to respond. â€Å"I want your permission to deviate from what we agreed to in our treaty with Ephraim. I want you to grant us an exception. I want your permission to save her life. You know 111 do it anyway, but I don't want to break faith with you if there is any way to avoid it. We never intended to go back on our word, and we don't do it lightly now. I want your understanding, Jacob, because you know exactly why we do this. I want the alliance between our families to survive when this is over.† I tried to swallow. Sam, I thought. It's Sam you want. â€Å"No. Sam's authority is assumed. It belongs to you. You'll never take it from him, but no one can rightfully agree to what I'm asking except for you† It's not my decision. â€Å"It is, Jacob, and you know it. Your word on this will condemn us or absolve us. Only you can give this to me.† can't think. I don't know. â€Å"We don't have much time.† He glanced back toward the house. No, there was no time. My few days had become a few hours. don't know. Let me think. Just give me a minute here, okay? â€Å"Yes.† I started walking to the house, and he followed. Crazy how easy it was, walking through the dark with a vampire right beside me. It didn't feel unsafe, or even uncomfortable, really. It felt like walking next to anybody. Well, anybody who smelled bad. There was a movement in the brush at the edge of the big lawn, and then a low whimper. Seth shrugged through the ferns and loped over to us. â€Å"Hey, kid,† I muttered. He dipped his head, and I patted his shoulder. â€Å"S'all cool,† I lied. â€Å"I'll tell you about it later. Sorry to take off on you like that.† He grinned at me. â€Å"Hey, tell your sister to back off now, okay? Enough.† Seth nodded once. I shoved against his shoulder this time. â€Å"Get back to work. I'll spell you in a bit.† Seth leaned against me, shoving back, and then he galloped into the trees. â€Å"He has one of the purest, sincerest, kindest minds I've ever heard,† Edward murmured when he was outof sight. â€Å"You're lucky to have his thoughts to share.† â€Å"I know that,†! grunted. We started toward the house, and both of our heads snapped up when we heard the sound of someone sucking through a straw. Edward was in a hurry then.He darted up the porch stairs and was gone. â€Å"Bella, love, Ithought you were sleeping,† I heard him say. â€Å"I'm sorry, I wouldn't have left.† â€Å"Don't worry. Ijust got so thirsty – it woke me up.It's a good thing Carlisle is bringing more. This kid is going to need it when he gets out of me.† â€Å"True. That's a good point.† â€Å"I wonder if he'll want anything else,† she mused. â€Å"I suppose we'll find out.† I walked through the door. Alice said, â€Å"Finally,† and Bella's eyes flashed to me. That infuriating, irresistible smile broke across her face for one second. Then it faltered, and her face fell. Her lips puckered, like she was trying not to cry. I wanted to punch Leah right in her stupid mouth. â€Å"Hey, Bells,† Isaid quickly. â€Å"How ya doing?† â€Å"I'm fine,† she said. â€Å"Big day today, huh? Lots of new stuff.† â€Å"You don't have to do that, Jacob.† â€Å"Don't know what you're talking about,† I said, going to sit on the arm of the sofa by her head. Edward had the floor there already. She gave me a reproachful look. â€Å"I'm so s – † she started to say. I pinched her lips together between my thumb and finger. â€Å"Jake,† she mumbled, trying to pull my hand away. Her attempt was so weak it was hard to believe that she was really trying. I shook my head. â€Å"You can talk when you're not being stupid.† â€Å"Fine,I won't say it,† it sounded like she mumbled. I pulled my hand away. â€Å"Sorry!† she finished quickly, and then grinned. I rolledmy eyes and then smiled back at her. When I stared into her eyes, I saw everything that I'd been looking for in the park. Tomorrow, she'd be someone else. But hopefully alive, and that was what counted, right? She'd look at me with the same eyes, sort of. Smile with the same lips, almost. She'd still know me better than anyone who didn't havefull access to the inside of my head. Leah might be an interesting companion, maybe even a true friend – someone who would stand up for me. But she wasn't my best friend the way thatBella was. Aside from the impossible love I felt for Bella, there was also that other bond, and it ran bone deep. Tomorrow, she'd be my enemy. Or she'd be myally. And, apparently, that distinction was up to me. I sighed. Fine!I thought, giving up the very last thing i had to give. It made me feel hollow. Go ahead. Save her. As Ephraim's heir, you have my permission, my word, that this will not violate the treaty. The others will just have to blame me. You were right – they can't deny that it's my right to agree to this. ‘Thank you.† Edward's whisper was low enough that Bella didn't hear anything. But the words were so fervent that, from the corner of my eye, I saw the other vampires turning to stare. â€Å"So,†Bella asked, working to be casual. â€Å"How was your day?† â€Å"Great. Went for a drive. Hung out in the park.† â€Å"Sounds nice.† â€Å"Sure, sure.† Suddenly, she made a face. â€Å"Rose?† she asked. I heard Blondie chuckle. â€Å"Again?† â€Å"I think I've drunk two gallons in the last hour,† Bella explained. Edward and I both got out of the way while Rosalie came to lift Bella from the couch and take her to the bathroom. â€Å"Can I walk?† Bella asked. â€Å"My legs are so stiff.† â€Å"Are you sure?† Edward asked. â€Å"Rose'llcatch me if I trip over my feet. Which could happen pretty easily, since I can't see them.† Rosalie set Bellacarefully on her feet, keeping her hands right atBella's shoulders. Bella stretched her arms out in front of her, wincing a little. â€Å"That feels good,† she sighed. â€Å"Ugh, but I'm huge.† She really was. Her stomach was its own continent. â€Å"One more day,† she said, and patted her stomach. I couldn't help the pain that shot through me in a sudden, stabbing burst, but I tried to keep it off my face. I could hide it for one more day, right? â€Å"All righty, then. Whoops – oh, no!† The cup Bella had left on the sofa tumbled to one side, the dark red blood spilling out onto the pale fabric. Automatically, though three other hands beat her there, Bella bent over, reaching out to catch it. There was the strangest, muffled ripping sound from the center of her body. â€Å"Oh!† she gasped. And then she went totally limp, slumping toward the floor. Rosalie caught her in the same instant, before she could fall. Edward was there, too, hands out, the mess on the sofa forgotten. â€Å"Bella?† he asked, and then his eyes unfocused, and panic shot across his features. A half second later,Bella screamed. Itwas not justa scream, it was a blood-curdling shriek of agony. The horrifying sound cut off with a gurgle, and her eyes rolled back into her head. Her body twitched, arched in Rosalie's arms, and then Bella vomited a fountain of blood.

Inclusion of the mothers from around the country was a cunning decision made by the managers at P&G the reason for this being was that mothers are the ones who usually purchase the products for the household or domestic use so convincing them and then make them work in marketing the new offering so an immaculate move by the management of P&G. Another benefit which P&G gained by devising this Vocalpoint program was that, housewives are hard to target as they do not use technology that often, rather they spend a big part of their leisure time in gossiping with their friends hence the best possible way to market the offerings to them was by this word of mouth publicity. P&G earned pretty much short term and long term benefits by applying this strategy; the most outstanding one was that launching this program helped the firm in increasing the sales by a considerable extent which obviously is the sole aim of launching any marketing campaign. In addition to this external clients of P&G also earned huge profits by using this program and so as a result P&G got its share in the profits, this amount was as higher as 1 million USD. The job of the R&D department got less hectic as it was able to get the desired feedback on the latest products; this was helpful for it in making changes to the existing products and also to the ones which are undergoing the process of development. One of the long term benefits which came to P&G’s way as a result of this program was that it got a solid loyal customer base on which it can capitalize in the forthcoming years. The ‘connect and develop† program of P&G also got immense support as a result of this program and the marketing intelligence of the company overall increased. The overall impact of this project on the position of the firm was immaculate as it was a win-win situation for the company; the managers were getting the ins and outs of the target market in no time whatsoever so the overall time consumed in the processes of product development got reduced. This decrease in the overall product development time acted as a competitive advantage for the managers as they were able to deliver the market with the products it wanted in much lesser time than the competitors. Another addition was that the managers at the firm were getting customer generated feedback on the latest products and that too in negligible time so this enabled them to alter their strategies if needed be; the consumer on the other hand were liking this as they were getting tailor-made products for their usage (Sirgy 1998). Answer2: Analyzing from the business perspective one has to say that the programs such as Vocalpoint and Tremor must go on as they are beneficial for the businesses in the longer run and same is the case with P&G. Having said that one has to admit that these projects and programs are certainly not the ones, which are based on the ethical principles and norms laid by the society. In my opinion there are various ethical issues which are present in such kind of WoM programs. The first and the foremost being is that the person to whom the offering is being marketed is unaware of the reality he is perceiving the interaction as a social one but in general the other person is acting as a sales person for a specific firm. My objection regarding this is that if a person is being marketed something then he must have prior knowledge of this. For me there is a thin line between marketing and deceiving and this difference has to be understood by the firms and the marketing managers; if there are ample ethical ways of marketing then what is the point in using the deceptive means? Another issue in my opinion is that by hiring or deploying WoM marketers the managers are making the society increasingly materialistic even the sacred relationships like friendship etc are being used just as an asset and even that without the other person being aware of this all. Objectifying the society is one thing which to me is intolerable and as this eventually ruins the whole basic structure of the civilization which really is denting in terms of the future perspectives. Considering the Tremor program I can firmly state that this was one project on which I have serious reservations; the reason for this being is that P&G was using minors (children under 18 years ) to market its products; this is one ridiculous way of marketing. The reason for this is that this is the age where the person learns the most, from its society and surroundings and if the society is indulged in inducing the materialistic thoughts in kids then this will be injurious for the entire social setup; this is the stage at which the children must be taught how to honor and value the relationships but such deceptive marketing projects destroys the whole brought up of a child. One legal issue which too arises as a result of the inclusion of minors in the marketing programs is that these kids are underage legally so they cannot be made a part of any marketing project without the prior consent of their guardians (Malachowski 2001); this is one thing which was completely neglected by the managers at P&G and hence they violated the rules and regulations laid by the government. In the Tremor programs the minors were not allowed to tell the reality to the person to which they are interacting this is something which is immoral and to me for this the firm has to be penalized; can we teach our younger generations to indulge into immoral activities just for the sake of money? This is a million dollar question which has to be answered by the managers at P&G. The Tremor program targeted the minors too which is also an unethical way of making business the reason for this is that the underage people are immature hence they cannot make decisions for themselves (Malachowski 2001); influencing a buyer’s decision by using rational marketing ploys is a justifiable act but making the kids addict by using deceptive means and backdoor channels has to be abandoned as this certainly is not favorable for the social composition. In sum commercialization of human relationships is one thing which must be stopped by the marketing managers if someone is indulging in WoM marketing of a firm then he should disclose his affiliation as this is beneficial for the society; involving money in the activities of daily life will hurt the society in the longer run. In the Vocalpoint program the managers at P&G were looking to select the mothers which are more social; the main reason behind this move was that, as mother is the most sacred and trustworthy relationship in this universe so mothers enjoy a unique position in the society; using their social stature for the marketing purposes was an unreasonable attempt. Answer 3:  The value of WoM is immense in the present global marketing scenario, the primary reason for this is that due to the increase in the globalization the nature of the competition has increased exponentially and so to do business and earn profits a firm has to market it’s offering to the target buyers. With the increase in the intensity of competition and technology the media of marketing have too increased and thus in such an overall marketing environment WoM publicity is something which is of great significance (Bothma 2003). In some cases ones counseling can prevent a negative life and provide for a safer society. This case depicts a client that falls under those circumstances. As a client, John Wayne Gacy will be introduced to Family Therapy to help him understand his family environment and help him to cope and or change the way he feels about his family life. The counseling plan will deal with John Wayne Gacy’s issues of his father being an alcoholic, his father abusing him and the rest of his family, him liking boys, as well as his anti-social ways at school. These will be addressed by looking at creating a genogram. When looking at the research it is favorable to use family therapy in order to facilitate change within the family environment by looking at problem solving abilities, looking at the positive rather than the negative, and accepting family members. Case Study My client is John Wayne Gacy, a 15 year old boy living in Chicago, Illinois. He is a white male and has been referred by a judge to undergo a psych evaluation, after his psych evaluation he began coming to see me. This is Gacy’s first time seeking help from a licensed counselor. Presenting Problems John’s presenting problems are his father is an abusive alcoholic, John is attracted to boys, and John is anti-social at school. These three things lead to much confusion in John’s life which led to him hearing voices. John wants to see if family therapy could change some of his issues and feels that by going to family therapy his home environment might change into a more supportive one. Family Background and Developmental History John Wayne Gacy, the son of Marion Elaine Robinson Gacy and John Wayne Gacy Sr. was born on March 17th 1942 in Chicago, Illinois. His parents were of Polish and Danish heritage (Bell). John was the middle child of three; he had an older sister Joanne and a younger sister Karen. John and his sisters were brought up Catholic and went to catholic school. The environment in which he was raised in was that of middle class, he had a part-time job after school and was involved with the boy scouts. John and his two sisters were raised by a father that was an alcoholic; he abused them and their mother. John was not popular in school and was often left out of the loop. John had a playground incident at age 11 which led to him having a blood clot and the clot caused him to have blackouts (Bell Bardsley). He could not play with the other kids because he had a heart condition which was another reason he was looked down upon by his father; he saw it as another shortcoming. John also discovered that he liked boys and often struggled with dealing with his sexuality (Bell). John’s father in a way resented him for this. He felt that it was a sign of weakness or another fault that John had. Current Living and Work Environment John is involved in many organizations within the community. He is a hard worker that often suffered from exhaustion and anxiety from the pressure he put on himself to succeed. John was a father and husband; and had a house in the suburbs. All of this came to a screeching halt when rumors of Gacy liking boys surfaced. Multiple charges and even a prison sentence were handed down to Gacy for the assault of a young boy. Physical Appearance, Mood, and Demeanor John was overweight and had heart and back problems; however, it did not stop him from his work or community activities. Gacy was sentenced to ten years in prison for assaulting a young boy. Gacy was in denial saying that the young boy willingly participated in the sexual acts in order to make extra money. He also did not take responsibility for his actions and tried to place blame on others. Gacy’s has an anti-social personality but is mentally competent. When he was in prison he followed rules and was not violent. Gacy took on different personalities as well as went through many different moods (Inside John Wayne Gacy’s Mind). Counseling Plan This counseling plan uses Family Therapy. â€Å"Families are systems of interconnected and interdependent individuals, none of whom can be understood in isolation from the system.†(Genopro) Basically family members feed off each other, positivity feeds positivity and vice versa. The best way to put together a treatment plan is to build your family genogram. Study your own behavior patterns and how they fit in with the rest of your family throughout generations. This will bring out options in which could be used in order to solve problems and figure out how one fits into the family and what ones role is within the family (Genopro). The genogram structure of family therapy will allow John and me to create a counseling plan very easily. Conceptualization of Client’s Presenting Problem and Developmental History John shows signs of multiple personality disorder as well as having an anti-social personality. John also hears voices. Family Therapy eludes to the fact that Family development and the environment that he was brought up in, along with family makeup could be a contributor to why John suffers from what he does (Capuzzi Gross, 2011). Family patterns or expectations are looked at in many ways from parental beliefs, personalities, and extended family influence. This allows each family to be unique in their own ways. This uniqueness can be very overwhelming and takes a toll on all members whether it is negative or positive (Pope). For example, John’s father puts a lot of pressure on him to be successful. John’s shortcomings were looked down upon by his father most likely do to the beliefs and personality that he formed based upon his parental upbringing. John admits that his father was abusive to his mom, sisters, and him as well. John took on many personalities and even called himself pogo the clown at times. John was attracted to boys and this is something that has haunted him his whole life and the reason he is in the predicament he is in. He hated being a disappointment to his father and he was always looking to please his father. John’s confusion with his sexuality ultimately lead to him killing those he sexually encountered because the voices he heard were telling him it would please his father. In session we will discuss with John  and his family ways in which the environment John is brought up in could be altered. Conceptualization of Human Nature According to Family Theory The goal of Family Therapy models is to take the problem presented by the individual dealing with their relationships with other people in the family setting and change the negative aspects (Franklin). The Family Therapy counselor believes the development of family life include family life cycle, family life spiral, and the family genogram (Capuzzi Gross, 2011) The family life spiral looks at how the members of the family go through the stages of development from young to old age. The family life spiral looks at generations and how they coincide with each other. The genogram charts development of generations. Counseling Goals The overall objective is for John Wayne Gacy to be able to find a way to cope with his anger towards his father and to be able to express how his father’s abuse towards the family affected him in a negative way. John said he wants to break down the barrier between him and his father because he loves him and ultimately wants to make him proud. To advocate for this John’s goal is to try and change the cognitive, affective, and behavioral aspects of his family system (Capuzzi Gross, 2011). In other words he wants to change the environment in the Gacy household completely around. The abuse needs to stop, the constant pressure needs to stop, and the pointing out of his weaknesses needs to be approached differently. Course of Treatment In order to get a complete understanding I believe the first action that needs to be taken is the nuclear family emotional system needs to be broken down, what this is looking at is the possible relationships with in a family and look at where problems could possibly come about. For John, 3 of the relationship patterns are touched upon in his situation. His father is abusive to his mother and siblings, in John’s fathers eyes there is an impairment of John because he likes boys, and lastly there is an emotional disturbance suffered by John because of his father’s view of him (Genpro). The second part that needs to be addressed is differentiation of self which is the dependency on others to give acceptance and approval. This is something John struggled with and never received from his father. He never felt like his father truly accepted him or approved of the life style that he lived (Genpro). John agreed to do 2 things a day to satisfy his needs, meaning if he wanted to work in the community do it because it made him feel good not because he wants or needs approval from his father. Treatment plan was developed and signed by John and we will meet on a weekly basis. Family Therapy Interventions Due to John stating that the pressure his father puts on him within the family system to be successful, the first intervention to be used will be the family interview. We will follow the five stages of the interview which are social, problem, interaction, goal setting, and ending (Capuzzi Gross, 2011). This interview will give me the tools necessary to properly assess the situation. The interview will also help John and his family understands how each other feels and come up with some interventions to help facilitate change. â€Å"the first obligation of a therapist is to change the presenting problem offered. If that is not accomplished, the therapy is a failure† (Capuzzi Gross, 2011). The first intervention needs to address the problem that John has presented, based upon the interview the interventions that will be implemented will be geared towards progressing with Johns presenting problem. Research Support The research that was obtained was very helpful in forming a counseling plan or treatment plan for this particular client. Family Therapy is known for helping individuals whose presenting problems deal with issues that stem from the family and the environment the individual was brought up in. Family therapy is the best way to look at â€Å"the patterns of communication and the relationships that connect people with one another and to their social and physical environments† (Capuzzi Gross, 2011, p. 287). In this case we will be looking at how John fits into his family and what his father’s expectations are of John. Interviewing as an intervention will be crucial in figuring out John place in the family and seeing how his and his father’s view of the roles conflict with each other. Family therapy allows for John  and his family to work on their problem solving with each other within the family, expressing their individuality with each other, discussing the positive and negative aspect of their family atmosphere as well as with each other on an individual basis, and lastly the acceptance of each other (Capuzzi Gross, 2011). Family therapy allows for clients to understand their environments as well as to understand their role within that environment. The counselor’s role within the session is to basically guide the discussion and conduct the interview. Family Therapy I feel is the best theory to use for a client such as John Wayne Gacy due to his confusion within his family, particularly when it comes to the relationship with his father and the expectation that his father has for him or at least how John see those expectations. Conclusion This counseling plan reviewed the use of Family Therapy for counseling John Wayne Gacy. Family Therapy is designed to be an extensive look at John Wayne Gacy’s family makeup. John Wayne Gacy’s presenting problems are his father abusing him and his family, his father being an alcoholic, he struggles with his sexuality, and he is anti-social. Family therapy is used to deal with these concerns, so for John, the counseling objective is to find a way to cope with his anger towards his father and to be able to express how his father’s abuse towards the family affected him in a negative way. The counseling plan addresses John’s concerns of worrying about his father’s abuse and constant push for perfection as well as his anti-social tendencies. Due the treatment plan and goals presented and the attitude of John to want to get better, I feel that Family Therapy will be successful for John Wayne Gacy as well as his family. A Teachers View On Teaching Kindergarten English Language Essay Kindergarten teachers establish the foundation of all education. These extraordinary educators help their students learn the alphabet, how to count, and how to write; all the qualities needed to be successful in school. In addition to academics, kindergarten teachers are instrumental in setting standards for good behavior in school. Without their guidance the children would not have the proper foundations for their future learning. Kindergarten classrooms and teachers are very important to little children who are just beginning their formal learning process at a new school. Kindergarten teachers are the most important in a childs education. Kindergarten is the second stage in school following pre-kindergarten. In pre-kindergarten you learn how to get along with and play with other students. Kindergarten is the establishment of young childrens existence in the educational system. What does the word kindergarten stand for? It means a school or class for children usually from four to six year olds à ¢Ã¢â€š ¬Ã‚ ¦ that serve as an introduction to school. (Merriam-Webster) Some teachers said it is not an easy job teaching kindergarten students. Two teachers interviewed say teaching kindergarten is a difficult job. Mrs. Van Dyke stated, Yes, yes, yes!! I really enjoy teaching them, but every day presents its own trials. It also has many rewards, (Mrs. Van Dyke). Mrs. Byrd stated, The good thing about kindergarten is the children are enthusiastic to learn and strive to please their teachers. I like being able to be the one that teaches the establishment of letters, phonics, numbers, and reading comprehension, (Mrs. Byrd). Mrs. Van Dyke and Mrs. Byrd where asked what inspired them to become a teacher? Mrs. Van Dyke said, I have always loved children. I began working as a dance teacher and enjoyed it so much I decided to go into it as a career, (Mrs. Van Dyke). Mrs. Byrd stated, I started working in the school system so that I could spend more time with my two sons. I worked three years in second grade. My Assistant principle moved me to kindergarten becau se she said my gift was really with the little ones, (Mrs. Byrd). Without the teachers the kindergarten class would not happen. The teachers will help the students to get all of the foundation needed to go on to the first grade. In essence, kindergarten teachers are very important to a little childs educational career. In an interview, Mrs. Van Dyke and Mrs. Byrd were asked how long it took them to complete their degrees to become a teacher and teacher assistant. What degree(s) did they get from college, and what college(s) did they attend? Mrs. Van Dyke stated, It took me four years. I took classes during the summer and graduated in four years. I attended Harding University and Slippery Rock College, receiving a degree in early childhood education, (Mrs. Van Dyke). Mrs. Byrd said, I have an associates degree in computer science from Gaston College. I worked full time in a textile mill so it took me between three to four years to complete it, (Mrs. Byrd). A classroom is a place for learning experiences. Most kindergarten classrooms have a kitchen play house set. Also the classroom contains water paint, a computer, a sand table, blocks, puzzles, and a writing center. These activities allow students to interact with each other students in the class. In a kindergarten classroom you have cubbies to store students supplies (book bag, clothes and other stuff). Also you will find a smiley chart which is used to display students behavior for the day. It is called a behavior chart; this chart is a scale showing a range of behaviors from high spirits to low or disappointing behavior from a teachers point of view. Lesson plans are a teaching tool to show how a teacher gets ready for his or her class every day. Lesson plans show what teachers do to help each and every student. Carpet time in kindergarten classes is a learning period. During carpet time the kindergarten students learn the days of the week. That is very important for little children to know the days of the week. Also during carpet time the children sing the good morning song. Here are the words to the song; Good morning to you! Good morning to you! Were all in our places with bright shining faces. Oh, this is the way to start a great day! Good morning to you! Good morning to you! It is very significant that the children know the days of the week, how to count, and read previous to first grade. Every now and then when the students are misbehaving they do not get to go outside and play with other children. That is called a time out. Some children do not like that at all. Also when some of the students misbehave they have to pull a clip. That is one way to tell when a student is being terrible. All of the children love to be interactive. [The little students will be devoted to this lesson because they are interactive in on the increase their class tree and then take an interactive piece in putting their letter on the tree when it is their turn to put their letter on the tree.] That is very full of life measurement in that days lesson plans. All most all of the kindergarteners are not able to sit in their sits all day long, they need to get up and be in motion around sometimes. If you have something in your lesson this is a good song to use. This is how the song goes like this. But its full again when we shipshape up at the end of every day.] That is one way to get their small bodies moving, and the kindergarteners will not be so wiggly when youre ready to be knowledgeable. In kindergarten the students may also have some home work to do. The students may have home work is really not difficult. At the start of the school year, the teachers will help the students learn how to say and write their alphabet. Some say that is too much on a little kindergartener. But it is really not. Students have to know the alphabet so when they get to first grade. They are the building blocks needed throughput their educational career. In addition to songs, nursery rhymes are another way to get the students active. Here are two examples of nursery rhymes, twinkle, and twinkle, and little star, and Hickory, dickory, dock. Hickory, dickory, docks; the mouse ram up the clock. The clock struck one, and the mouse ran down the clock, Hickory, dickory, dock (Mrs. Byrd). The alphabet comforter is a good thing to use because it refreshes their memory allowing them to remember their sounds and how to pronounce them; some teachers say this lesson is great for little ones. To help the students count, the teachers can use some number word rhymes to help them remember how to count. The teachers also can help them to remember their colors by doing color rhymes or songs that tells them how to spell the colors of the rainbow. Here is one of the color rhymes for the color black. Another method of learning is where the teacher begins by trying to activate prior comprehension is. The teachers questions will help them to understand what they will be learning. The teacher will encourage students to start developing skills in listening to others, sharing thoughts or a dream, thinking about what they know, thinking about what they would like to know, and following directions. The teachers will also assist students in developing a positive reception for other ideas. A positive reception for the topic they will be studying. A feeling of accomplishment. Core curriculum content standards; all students will get your hands on historical understanding of societal thoughts and forces throughout the history of New Jersey, the United States, and world. Introduction; review; the teacher will ask the students what they are on familiar terms with about communication. Enthusiasm the teacher will have books, fiction and nonfiction, displayed on the topic. Predicament what so we already know about communication and what do we want to know? Development; the teacher will tell the class that they will be studying about communication. Ask the class what they think communication is. The teacher will ask the class what types of things they would like to be familiar terms with about communication. The teacher will go over the responses he or she has written on the paper and put it into simple words. The teacher will explain the different things they will be learning with reference to ways communication happen, inventions, a nimal communication by Aliki, graphic representation document. Some students may or may not like the way the teacher teaches, that is very okay. This is why kindergarten teachers and classroom in a childs educational career. Life as a kindergarten teacher can be very hard in a way. The teachers have a very big job on their hands because teaching kindergarten is hard knowing what that child goes through at home. Some parents just do not care about their childs school work. So with that being said it is very hard sometimes to be a teacher. The teachers will never know how a child is treated at home unless they tell you. So kindergarten teachers are so very important in a childs educational career. It is exceptionally okay that a child goes to pre kindergarten taking place before he or she goes off into kindergarten to establish on his or her educational career. Graduation Speech: Perseverance and Determination :: Graduation Speech, Commencement Address

As a hurdler for County High, I stumble upon some pretty tough obstacles each time I race; the same is true in life. Everyone encounters obstacles, but in order to overcome them and succeed, one must never give up. I love to run hurdles, but unfortunately last year, little pulls and strains prevented me from running to my full potential. One Thursday, we had a home track meet against Lake Stevens. For the first time I was in pretty good shape for my race, the 100-meter hurdles. I began jumping up and down partially to stay warm, and partially to let out some of my excitement. By this time, I had butterflies in my stomach and the adrenaline was pumping. The starter asked us to 'Take your sweats off and stand behind your blocks.' 'Runners take your marks.' Hands shaking, I crouched into the starting blocks. The gun was up. 'Set!' 'Bang!' I bolted out of the blocks. I was way ahead of the other girls when suddenly, I realized I didn't have enough speed to carry me over the next hurdle. Gathering all of the strength I could, I grabbed at the air in hopes of guaranteeing clearance. I had just brushed over the wood when my foot hit the ground and my ankle gave out. I fell. I heard a gasp from the cro wd and the other racers' feet pounding past me. I got back up. I had never gone over a hurdle with my right leg first, but I did after that fall. Sprinting as fast as I could in between hurdles, I found myself basically bunny hopping over the rest of them. My goal was to cross that finish line and to be able to say that I did the very best that I could, even if I didn't look very graceful along the way. Although it might have seemed like a bad day, I was proud. It was the first time I had ever fallen in a race, and not only did I get back up and keep running, I managed to place second. Sometimes you encounter hurdles in life - sometimes you have to adapt and alter your normal routine to clear them. Then there are times you fall; the people surrounding you may gasp - they had been rooting for you. But they haven't given up on you-it's now more than ever that they want you to succeed - it's human nature to want the underdog to come from behind.

Creativity The walls of the Louvre are covered with artistic masterpieces, widely recognized as some of the most treasured, creative works known to man. The unique stroke of a brush or the layering of paint can distinguish these geniuses from the masses. Despite the ability to label these pieces of art as especially creative, it remains unclear how one can truly qualify the defining characteristics of creativity. This faculty is commonly viewed as intangible, as `a spark of creativity,' or, `a flash of creativity.' This phenomenon, which lies at the foundation of our artistic culture, eludes our current grasp of understanding. For some, creativity rests within our human nature as an integral part of our being, for others, it may emerge from the great mechanical complexity of our brains, or the randomness of the universe. In recent years there have also emerged visual and literary works by computers which some experts believe to have a creative nature. This controversial idea causes unease in ma ny that characteristics we commonly assume to be innately human could manifest in a machine. However, after viewing the works created by computers, it becomes necessary to consider the possibility of such creative ability whether or not the works themselves are creative. Though one can find examples of human creativity in nearly all aspects of life, perhaps it is most clearly evident in our literature. The thoughtful combination of words can express the gauntlet of human emotions and experiences aptly, and in the most talented of authors, can breathe life into the words and seemingly recreate the experiences in the mind of the reader. Notable and critically praised author James Joyce provides an example of this in his work Ulysses: .. ...uture computer which could paint like Monet, or have faith in a machine to effectively speak to the emotions of a human. However, technological innovations continually astound us, and it is no less plausible that a computer could function at this level of creativity, than one hundred years ago to imagine men landing on the moon. Rachel Reed PHIL 389 Saint-Croix 10 February 2014 Question #7: Descartes’ Foundationalism In this paper I will describe the foundationalist structure of Descartes’ arguments in his work Meditations on First Philosophy. Foundationalism is the view that there are some beliefs are epistemologically basic and can be known without knowing anything else is true (Loeb, Lecture 1-14). For example, philosophers such as Descartes would acknowledge that geometric truths, such as 2 + 2 = 4, are so fundamental that they don’t need to be proven through argumentation. Thus, these truths can provide the basic foundation for further arguments. In my paper, I will show that two foundational claims of Descartes are first, the existence of the mind, and second, the existence of God. From these claims Descartes derives many others, including the argument for material objects and souls. As I lay out Descartes’ case, I will examine the philosophical soundness and validity of his foundationalist account, as well as its merits and potential weaknesses. In the end, I will conc lude that Descartes’ foundationalism, while alluring in its simplicity, does not survive deeper investigation. Descartes’ first foundational argument asserts that one can have knowledge of one’s own existence. The claim is essential to many arguments that follow because it survives his â€Å"Deceiver Hypothesis.† This hypothesis states that â€Å"there may be a powerful deceiver of supreme power who is deliberately and constantly deceiving me† (Med III, p. 17). This demonstrates that we cannot know, or be sure of, anything based on sensory experience alone. However, Descartes supports the idea that some things can be known entirely outside of sensory experience; through the use of logic and re... ...undational premises, such as the existence of God and the mind, do not provide indisputable groundwork for Descartes’ argument. First begging the question to prove the existence of the mind via dualism, and then conflating logic with cultural and personal ideals, these two tenets cannot stand on their own. Quantification of Proteins in Solution by Spectrophotometer Essay

Introduction: Absorption spectroscopy is a common method for finding the concentration of proteins or protein complexes in a solution. Proteins absorb light at specific wavelengths and can be defined by the equation A = log (Io/I). This equation states that an absorbance at a specific wavelength, A is equal to the log of the ratio of incident light intensity (Io), to transmitted light intensity (I). A spectrophotometer can be used quantitatively and qualitatively. A spectrophotometer is used qualitatively to obtain an absorption spectrum, which can be obtained by plotting the absorbance values, over the range of wavelengths tested for the solution. This helps to find out the suitable wavelength that the compound absorbs maximum. And the spectrophotometer is used quantitatively by using the Beer-Lambert Law; Log [ Io/I] = A = ÃŽ ¾cl, where ÃŽ ¾ is the molar extinction coefficient (unit = Lmol-1cm-1), helps to define the absorbance of the protein, c is the concentration of the substance (mol liter-1), and l is the path length of the light (unit = cm) through the medium. Log [Io/I] is called optical density or absorbance of the substance, and does not have units. Also, an absorption spectrum is created, which deals with absorption and wavelength (nm) of light used, with which â€Å"maximum absorption† is observed. Maximum absorption is when most of the solution particles are absorbed, and this happens at a specific wavelength. Since the Beer Lambert law is useful only for a range of wavelengths, it is not applicable to all protein solutions. In this experiment, an absolute standard was calculated using BSA, so that the concentrations of the other unknown protein solutions can be determined (Lambert et.al, 2011).The different assays used for this protein quantification were Lowry, Bradford (Coomassie Blue) and UV direct. Protein assays help to determine the amount of desired particle present (Srivastava, 2008). The aim of this lab is to understand the various aspects of spectrophotometry and its applications in biochemistry, such as quanitification of protein solutions. Absorbance is noted to increase when wavelength increases till it reaches the point of maximum absorption, after which it decreases till it nearly reaches zero. It is best to consider wavelength of maximum absorption because stronger the intensity, the more accurate will be the readings for absorbance. As seen from table 1, the path lengths remain the same as the cuvettes used were of the same size. The Beer-Lambert Law states that Abs = Æ .c.l, where Æ  = molar extinction coefficient, c = concentration of protein solution, and l = path length of light through medium. Thus, it is noted that absorbance and path length share a directly proportional relationship, i.e. if path length increases, absorbance increases as well. It was clear ly observed in the wide and narrow test-tubes, that as the path length was doubled, the absorbance value doubled too (Srivastava, 2008). Also, from the same equation, it can be determined that absorbance and concentration share a directly proportional relationship meaning that as the concentration decreases, it directly affects the absorbance value obtained, and this value decreases too. Thus, as seen for the four cuvettes tested (in Table 1) as the concentration is halved in every cuvette, the absorbance value is halved correspondingly as well. It is known that the Beer-Lambert law says absorbance is proportional to number of absorbing molecules, and that this is valid for a variety of compounds over a wide range of concentrations. But even as the molar extinction coefficient is seen to be attributed to wavelength, it is true only for monochromatic light (Lambert et.al, 2011). The relationship can be stated as â€Å"Æ  is a measure of the amount of light absorbed per unit concentration†. Molar extinction coefficient is a constant for a particular substance, therefore according to the Beer-Lambert Law it is expected that if the concentration of the solution is halved so is the absorbance. A compound with a high molar extinction is very effective at absorbing light (of the appropriate wavelength), and hence low concentrations of a compound with a high molar extinction can be easily detected. In the values determined (Table 1), the experimental values are in accordance with the theoretical statement except for one cuvette. The cuvette no.3 with Æ  = 1.8Ãâ€"10-4 L mol-1cm-1 does not agree with the trend. Thus it can be deduced that due to experimental error, the Æ  value is inaccurate. Also, from the equation it is understood that Æ  and path-length are inversely proportional as well (i.e. Æ  = Abs/cl) that means that as path-length increases, Æ  decreases, assuming that the concentration is kept constant. But the experimental values do not agree with this statement, because it is seen that as the path-length increases so does the molar extinction coefficient, Æ . Biochemical methods are applied for to determine protein concentration in solutions. Many techniques are less used because they have limitations such as reduced sensitivity, time available for the assay, or they are highly specific about the amino acids in the protein solution being tested. But for e very protein, the component amino acids are different, so there is no single assay that can be used for quantification of all proteins. The absorbance assays use the method of testing the intensity of the color produced by the protein solutions when chemical reagents are added to it. A â€Å"standard protein† whose concentration is known, is treated using the same chemical reagents and thus an absolute standard curve is obtained (Boyer, 2000). In this experiment, the standard used was Bovine Serum Albumin (BSA). Development of color is significantly better in BSA than any other protein, and this makes it one of the most preferred test solutions for quantification of proteins (Antharavally et.al, 2008). Hence figure 2 is obtained by performing the three suitable assays on BSA to produce a standard curve, also it can be noted that only the UV direct gave a straight line passing through zero, whereas the Lowry and Coomassie Blue gave curved lines, passing through zero. Table 2 shows the absorbance values recorded, for different dilutions of the test protein in three different assays. With the help of the values obtained in Table 2, and with the equations obtained from Figure 2, the concentration of protein (mg/ml) was calculated and presented in Table 3. Since all the values in Table 3 were deduced from the equation of standard curve BSA, it is considered as the absolute standard, and the other test protein solutions are known as the relative standards. Using the values from Table 3, taking BSA as the absolute standard, the almost actual concentration of the protein (mg/ml) can be concluded, and they are 1.6 (mg/ml) for Lowry assay, 1.4 (mg/ml) for Coomassie Blue and 1.3 (mg/ml) for UV direct. For Lowry assay, the concentration value for all test proteins was 1.6 mg/ml, which must mean that the value obtained is accurate. For Coomassie blue, BSA and Hemoglobin were the same (1.4mg/ml), Ovalbumin and Lysozyme had similar values of 1.9mg/ml, and 1.8mg/ml respectively, whereas Gamma globulin showed 2.5mg/ml. The value for Gamma globulin is off because of experimental error, of spilling some of the contents from the cuvette while transferring it to the spectrophotometer for calibration. For UV direct, BSA and Ovalbumin have similar readings (1.3mg/ml and 1.5mg/ml respectively), Gamma globulin is 2.5mg/ml, but Lysozyme is 5.9mg/ml and Hemoglobin is 3.8mg/ml. The reason for this could be due to the fact that UV direct helps to identify the presence of aromatic compounds indicating that Lysozyme and Hemoglobin contain aromatic compounds present in them. The Lowry protein assay is the most common and one of the more sensitive, but it is time consuming, on the other hand Coomassie blue (the Bradford assay) is much more sensitive as compared to Lowry, and requires less time too. They both show change of color with proteins. As for UV direct method, it is one of the faster methods too, and it is helpful to identify aromatic compounds because aromatic residues absorb 280nm light (Boyer, 2000). The Lowry procedure can detect protein levels as low as 5 µg (Boyer, 2000). It depends on the color development by the reagent Folin-Ciocalteu. Peptide bonds are formed under alkaline Cu2+ conditions and reduced from Folin-Ciocalteu phosphomolybdate-phosphotungsten by aromatic amino acids (tyrosine and tryptophan) to heteropolymolybdenum blue. The standard curve obtained with BSA helps to determine concentration of unknown protein solutions (Antharapally et.al, 2008). In the case of Coomassie blue, it is more efficient than Lowry because even though there is variation with different proteins, there is very less interference by non protein components (Borley, 2000). Therefore, according to literature, Coomassie Blue is the most preferred protein assay but this contrasts the experimental inferences, because through experimental procedure it was seen that Lowry method gave the most accurate and precise results. With this experiment, the method to quantify unknown protein concentrations has been understood. Also, that this process must be performed carefully to avoid irrational experimental errors.
